چکیده

Oleuropein, one of the main polyphenolic constituents olive, is cardioprotective against ischemia reperfusion injury (IRI). We aimed to assess cardioprotection afforded by acute administration oleuropein and evaluate underlying mechanism. Importantly, since antioxidant therapies have yielded inconclusive results in attenuating IRI-induced damage on top conditioning strategies, we investigated whether could enhance or imbed manifestation ischemic postconditioning (PostC). given during as a single intravenous bolus dose reduced infarct size compared control group both rabbits mice subjected myocardial IRI. None inhibitors pathways, l-NAME, wortmannin AG490, influence its limiting effects. Combined PostC cause further limitation comparison with alone animal models. Oleuropein did not inhibit calcium induced mitochondrial permeability transition pore opening isolated mitochondria increase cGMP production. To provide insights different mechanism oleuropein, sought characterize anti-inflammatory potential vivo. their combination reduce inflammatory monocytes infiltration into heart circulating monocyte cell population. Oleuropein's action involves direct protective effect cardiomyocytes it significantly increased viability following simulated IRI non-treated cells. ?leuropein confers additive PostC, via increasing expression transcription factor Nrf-2 downstream targets In conclusion, provides robust synergistic experimental models inducing defense genes through axis independently classic signaling pathways (RISK, cGMP/PKG, SAFE).

The effect of oleuropein on the anxiety caused by acute stress via reducing oxidative stress in mice

Introduction: Severe stressful events are associated with the risk of mental disorders such as depression and anxiety. Due to the negative effects of anti-anxiety drugs, researcher’s attention have been focused on finding natural ingredients. The aim of the present study was to investigate the effect of oleuropein on anxiety induced by acute stress in male rats. Materials and Methodes: To induc...

Inhibition of myocardial injury by ischemic postconditioning during reperfusion: comparison with ischemic preconditioning.

Ischemic preconditioning (Pre-con) is an adaptive response triggered by a brief ischemia applied before a prolonged coronary occlusion. We tested the hypothesis that repetitive ischemia applied during early reperfusion, i.e., postconditioning (Post-con), is cardio-protective by attenuating reperfusion injury. In anesthetized open-chest dogs, the left anterior descending artery (LAD) was occlude...

Effect of oleuropein on myocardial dysfunction and oxidative stress induced by ischemic-reperfusion injury in isolated rat heart

BACKGROUND Studies have reported antioxidant effect of oleuropein in isolated rat heart. OBJECTIVE This study was conducted to investigate whether perfusion of isolated rat heart with oleuropein, before induction of ischemia or at the onset of reperfusion, had any effect on the hemodynamic parameters, infarct size and biochemical factors following ischemic - reperfusion injury. MATERIALS AN...

Combined Preconditioning and Postconditioning Provides Synergistic Protection against Liver Ischemic Reperfusion Injury

Hepatic Ischemia and Reperfusion Injury (IRI) is a major cause of liver damage during liver surgery and transplantation. Ischemic preconditioning and postconditioning are strategies that can reduce IRI. In this study, different combined types of pre- and postconditioning procedures were tested in a murine warm hepatic IRI model to evaluate their protective effects. Proanthocyanidins derived fro...
